چکیده

Fluorigenic substrates prepared from 4-methylumbelliferone provide a simple, convenient method for detecting and assaying group-specific hydrolase activity in small quantities of whole mycobacteria. Physico-chemical properties of the enzymes such as pH dependence and heat stability may also be studied. A technique is described for studying glycosidase, hexosaminidase, acid phosphatase, and acyl esterase activity in seven mycobacterial species. Such techniques will be a useful aid to the taxonomy, identification, and quantification of mycobacteria, especially slowly growing strains in which biochemical properties are very difficult to detect by other techniques.

Characterization of Leishmania Species and L. major Strains in Different Endemic Areas of Cutaneous Leishmaniasis in Iran

Both zoonotic and anthroponotic cutaneous leishmaniasis (CL) caused by L. major and L. tropica, respectively, are endemic in different parts of Iran. This study was performed to investigate the new changes in epidemiological pattern of CL, and to identify the species of Leishmania and the strains of L. major isolated from different endemic areas of Iran. Seventy-two isolates from 245 samples co...

Production of four Neurospora crassa lytic polysaccharide monooxygenases in Pichia pastoris monitored by a fluorimetric assay

UNLABELLED BACKGROUND Recent studies demonstrate that enzymes from the glycosyl hydrolase family 61 (GH61) show lytic polysaccharide monooxygenase (PMO) activity. Together with cellobiose dehydrogenase (CDH) an enzymatic system capable of oxidative cellulose cleavage is formed, which increases the efficiency of cellulases and put PMOs at focus of biofuel research. Large amounts of purified P...

A fluorimetric assay for the spontaneous release of an N7-alkylguanine residue from duplex DNA.

A fluorimetric assay for monitoring depurination of the N7-alkylguanine adduct derived from the anticancer natural product leinamycin is described. This general approach could potentially provide the foundation for a high throughput assay that detects DNA-alkylating agents or a convenient continuous fluorimetric assay for base excision repair enzymes.

Differentiation of M. scrofulaceum from Related Mycobacterial Species by Double-Diffusion Technique

Isolation and differentiation of various species of mycobacteria are predominantly based on conventional, cultural and biochemical tests. Among the non-tuberculous mycobacteria, these tests are unable to differentiate M. scrofulaceum from M. avium-intracellulare (MAI) because of their identical biochemical characteristics. For this reason, one hundred mycobacterial strains suspected to be M. ...

Fluorimetric coupled enzyme assay for lysoplasmalogenase activity in liver.

We developed a continuous spectrofluorimetric assay of lysoplasmalogenase activity with the use of horse liver alcohol dehydrogenase as a coupling enzyme. In this method the disappearance of NADH is measured spectrofluorimetrically. The excitation and emission monochromators were set at 340 and 460 nm respectively. The assay is 10 times as sensitive as the previous u.v. spectrophotometric metho...
